Isolated Plin5-deficient cardiomyocytes store less lipid droplets than normal, but without increased sensitivity to hypoxia

چکیده

Plin5 is abundantly expressed in the heart where it binds to lipid droplets (LDs) and facilitates physical interaction between LDs mitochondria. We isolated cardiomyocytes from adult Plin5+/+ Plin5-/- mice study role of for fatty acid uptake, LD accumulation, oxidation, tolerance hypoxia. Cardiomyocytes cultured with oleic stored less than Plin5+/+, but comparable levels when adipose triglyceride lipase activity was inhibited. The ability oxidize acids into CO2 similar cardiomyocytes, had a transient increase intracellular oxidation intermediates. After pre-incubation acids, retained higher content glycogen showed improved hypoxia compared Plin5+/+. In isolated, perfused hearts, deletion no important effect on ventricular pressures or infarct size after ischemia. Old reduced cardiac triacylglycerides, increased weight, apart modest elevated expression mRNAs beta myosin heavy chain Myh7 transporter Cd36, other genes involved metabolism glucose utilization were essentially unchanged by removal Plin5. seems facilitate storage primarily repressing without altering capacity. Expression has little importance acute ischemia, which contrasts protective mouse models during myocardial

Inhibition of ATR leads to increased sensitivity to hypoxia/reoxygenation.

The transient opening and closing of tumor vasculature result in periods of severe oxygen deprivation (hypoxia) followed by reoxygenation. This exerts a positive selective pressure for cells that have lost their sensitivity to killing by reduced oxygen levels. These cells are effectively resistant to hypoxia-induced apoptosis and conventional therapeutic approaches. Here we show hypoxia-induced...

Hydrolysis of lipid droplets in acid cholesteryl-esterase-deficient fibroblasts.

The mechanisms of hydrolysis and accumulation of cholesteryl oleate-lipid droplets prepared in vitro were studied in acid cholesteryl-esterase-deficient fibroblasts (GM00863, GM03111). Acid cholesteryl esterase activity was reduced in both GM00863 and GM03111 (8.9% and 17.4% of the normal level, respectively), while neutral cholesteryl esterase activity was highly stimulated in GM03111. The hyd...

Plin5 alleviates myocardial ischaemia/reperfusion injury by reducing oxidative stress through inhibiting the lipolysis of lipid droplets

Myocardial ischaemia-reperfusion (I/R) injury is a complex pathophysiological process. Current research has suggested that energy metabolism disorders, of which the abnormal consumption of fatty acids is closely related, compose the main pathological basis for myocardial I/R injury. Lipid droplets (LD) are critical regulators of lipid metabolism by LD-associated proteins. Among the lipid drople...

Lipid droplets

OK, they are cute, but why should a developmental biologist care about them? Onset of calcification early in ontogeny coupled with gradual, progressive morphological development permits the recognition of growth sequences for over a hundred trilobite species. Striking variation in the number of trunk segments at maturity across ontogeny and phylogeny, coupled with the curious pattern of segment...

XBP-1-deficient plasmablasts show normal protein folding but altered glycosylation and lipid synthesis.

The accumulation of misfolded secreted IgM in the endoplasmic reticulum (ER) of X-box binding protein 1 (XBP-1)-deficient B cells has been held responsible for the inability of such cells to yield plasma cells, through the failure to mount a proper unfolded protein response. LPS-stimulated B cells incapable of secreting IgM still activate the XBP-1 axis normally, as follows: XBP-1 is turned on ...
